Rapid Purification of Drug Enantiomers using Supercritical Fluid Chromatographic Method: Ibuprofen as a Model Compound

نویسندگان

چکیده

Abstract: Background: The purification of drug enantiomers is an essential technique because pharmacological activity differs depending on individual enantiomers. Nowadays, supercritical fluid technology utilized for various purposes in the pharmaceutical field. Methodology: In this study, efficient and streamlined method development strategy finding effective analytical chromatography (SFC) presented detail scaling up to preparative scale achieving enantiomerically pure product, using ibuprofen as a model compound. Results: Through optimized presented, (R) enantiomer commercial racemate can be obtained with recovery yield 80.0% enantiomeric excess 95.1 (S) 77.6% 99.3, respectively. Conclusion: use SFC systems has advantages over conventional terms separation time, reduced solvent consumption high productivity ibuprofen's purification. Separation of the Enantiomers of Ibuprofen by a Gas Chromatographic– Mass Spectrometric Method

Separation of the enantiomers of chiral drugs has become an important issue in analytical clinical chemistry in recent years, because of differences in the biological activity and pharmacokinetic properties of drug enantiomers [1,2]. Many drugs contain chirality centers; their enantiomers may have considerably different potency, type of pharmacological activity, pharmacokinetics, and metabolism...

Liquid-chromatographic assay of ibuprofen enantiomers in plasma.

This stereospecific "high-performance" liquid-chromatographic (HPLC) assay is suitable for pharmacokinetic studies of ibuprofen (IB). Very efficient extraction of the drug and internal standard, (+/-)-2-(4-benzoylphenyl)butyric acid, from plasma with isooctane/isopropanol (95/5, by vol) is followed by sequential reaction of the enantiomers with ethyl chloroformate and (S)-(-)-1-(1-naphthyl)ethy...
